The PathHunter CRHR1 Activated GPCR internalization cell line is a stable, engineered cell line for use in studying drug candidates (small molecules or biologics) affecting GPCR activation and internalization. This cell line uses EFC technology to measure ligand-induced internalization of CRHR1 via detection of localization to the early endosome. The included cell line overexpresses EA-tagged β-Arrestin-2 as well as a ProLink-tagged early endosome protein and untagged CRHR1. Upon ligand-induced receptor activation, β-Arrestin-2 is recruited to the activated GPCR and the resulting CRHR1-arrestin complex is internalized to the early endosome, leading to production of EFC signal. MC3T3-E1 Subclone 4 preosteoblasts were isolated from the cloned but phenotypically heterogeneous MC3T3-E1 cell line. This subclone was selected for high osteoblast differentiation and mineralization after growth in medium containing ascorbic acid. This cell line is a good model for studying in vitro osteoblast differentiation, particularly ECM signaling. It's behavior is similar to primary calvarial osteoblasts.

The PathHunter CXCR6 cell line is a stable, engineered cell line for use in studying drug candidates (small molecules or biologics) targeting GPCRs. This cell line enables assessment of ligand (e.g. CXCL16) based activation of CXCR6 GPCR activity via detection of β-arrestin recruitment. The included cell line overexpresses ProLink-tagged CXCR6 and EA-tagged β-Arrestin-2. Activation of CXCR6 stimulates the recruitment of β-Arrestin-2 and produces EFC signal.
